Humphrey was listed in a manor court record in 1629,
at the death of John Cowper. John was a hereditary tenant in the town of Holland in Duffield. At his death his estate owed a heriot to the lord of the manor. A heriot was a tribute owed to the lord of the manor, usually a farm animal. Humphrey was named as the son and heir of John's tenancy: "Humfrus Cowper est euis filius et here". The document was written in Latin.

Humphrey was named in another court record in Latin in 1629, along with his sons George, John, and Richard: "George Cowper filius" , "Johis Cowper filius," and "Ricd Cowper filius". The document seems to be a transfer of the land and cottage in Holland, inherited from Humphry's father John a year earlier, to George. The document also named Humphrey's wife Elizabeth: "Ad hoc curiam venit Humphrus Cowper et Elizabeth ux" - To this court came Humphrey Cowper and Elizabeth his wife.
